You do not need any license to start initially. But to continue getting updates, you should take the subscriptions. Otherwise, you will not be able to update SP Page Builder and EasyStore other than quickstart versions.

If/when I add the license, since I bought the personal for $99, and the Business with EasyStore is $249, do I pay the $249 total, or do I pay the difference between the two?
